Construction and humoral immune response of Epstein-Barr virus latent membrane protein 2 DNA vaccine

摘要: We constructed a eukaryotic expression plasmid encoding Epstein-Barr virus latent membrane protein 2 (EBV, LMP2) and evaluated its effects on humoral immunity. First, the encoding sequence of the EBV was amplified from B95−8 cell RNA by reverse transcription polymerase chain reaction (RT-PCR) and then was directionally cloned into eukaryotic expression vector pcDNA3.1. It was employed to evaluate immune response of the mice inoculated doubly with the DNA vaccine. The serum antibody against LMP2 was detected with enzyme-linked immunosorbent assay (ELISA). The recombinant plasmid pcDNA3.1- was confirmed by the restrictive endonuclease analysis and sequence analysis. The serum titer of IgG antibody against LMP2 epitope in the mice immunized with the DNA vaccine encoding LMP2 was up to 1∶4000. In conclusion, the EBV DNA vaccine can induce a strong humoral immune response in mice.

Effect of protein on PVDF ultrafiltration membrane fouling behavior under different pH conditions: interface

摘要: pH values of the BSA solution significantly impact the process of membrane fouling. Dramatic flux decline is caused by membrane–BSA adhesion force at start of filtration. XDLVO theory shows the polar or Lewis acid–base interaction plays a major role in membrane fouling. To further determine the fouling behavior of bovine serum albumin (BSA) on different hydrophilic PVDF ultrafiltration (UF) membranes over a range of pH values, self-made atomic force microscopy (AFM) colloidal probes were used to detect the adhesion forces of membrane–BSA and BSA–BSA, respectively. Results showed that the membrane–BSA adhesion interaction was stronger than the BSA–BSA adhesion interaction, and the adhesion force between BSA–BSA-fouled PVDF/PVA membranes was similar to that between BSA–BSA-fouled PVDF/PVP membranes, which indicated that the fouling was mainly caused by the adhesion interaction between membrane and BSA. At the same pH condition, the PVDF/PVA membrane–BSA adhesion force was smaller than that of PVDF/PVP membrane–BSA, which illustrated that the more hydrophilic the membrane was, the better antifouling ability it had. The extended Derjaguin–Landau–Verwey–Overbeek (XDLVO) theory predicts that the polar or Lewis acid–base (AB) interaction played a dominant role in the interfacial free energy of membrane–BSA and BSA–BSA that can be affected by pH. For the same membrane, the pH values of a BSA solution can have a significant impact on the process of membrane fouling by changing the AB component of free energy.

Protein phosphatase 2A, a key player in Alzheimer’s disease

摘要: Protein phosphatase 2A (PP2A) is the predominant serine/threonine phosphatase in eukaryotic cells. In the brains of patients with Alzheimer’s disease (AD), decreased PP2A activities were observed, which is suggested to be involved in neurofibrillary tangle (NFT) formation, disturbed amyloid precursor protein (APP) secretion and neurodegeneration in AD brain. Based on our research and other previous findings, decreased PP2Ac level, decreased PP2A holoenzyme composition, increased level of PP2A inhibitors, increased PP2Ac Leu309 demethylation and Tyr307 phosphorylation underlie PP2A inactivation in AD. β-amyloid (Aβ) over-production, estrogen deficiency and impaired homocysteine metabolism are the possible up-stream factors that inactivate PP2A in AD neurons. Further studies are required to disclose the role of PP2A in Alzheimer’s disease.

Dual faces of SH2-containing protein-tyrosine phosphatase

摘要:

PTPN11, which encodes tyrosine phosphatase Shp2, is a critical gene mediating cellular responses to hormones and cytokines. Against original prediction as tumor suppressor for tyrosine phosphatases, PTPN11 was first identified as a proto-oncogene because activating mutations of this gene are associated with leukemogenesis. However, most recent experimental data suggest PTPN11/Shp2 acting as a tumor suppressor in hepatocarcinogenesis. This review focuses on the tumor-promoting or suppressing roles of the gene PTPN11/Shp2 in different cell types.

Blockage of receptor-interacting protein 2 expression by small interfering RNA in murine macrophages

摘要: This study aims to demonstrate that blocking the receptor-interacting protein2 (Rip2) expression can decrease inflammatory cytokine production by macrophage and protect mice from endotoxin lethality. Murine Rip2 small interfering RNA (siRNA) plasmids were constructed and transfected into macrophage and Rip2 expression was detected with reverse transcription-polymerase chain reaction (RT-PCR) and western blot. Cell proliferation was assayed with MTT. TNF-? concentration was assayed with ELISA and high-mobility group box 1 protein (HMGB1) level with semi-quantitative western blot after lipopolysaccharide (LPS) stimulation. LPS challenge was given after the plasmids were injected into mice and the survival rate was calculated. Rip2 siRNA plasmid could block the mRNA and protein expression of Rip2 and promote cell proliferation. Blocking Rip2 could attenuate LPS-induced TNF-? and HMGB1 production. The HMGB1 expression in the liver decreased to (40.21 ± 11.03) pg/g, and serum TNF-? level decreased to (300.43 ± 59.26) ng/L ( < 0.05). The survival rate of mice from endotoxemia was also improved ( < 0.05). The results demonstrate that Rip2 siRNA plasmid can block the expression of Rip2, decrease the production of TNF-? and HMGB1 and protect mice from fatal endotoxemia.

Application of ultra-sonication, acid precipitation and membrane filtration for co-recovery of protein

摘要: A novel method was applied to co-recover proteins and humic acid from the dewatered sewage sludge for liquid fertilizer and animal feed. The proteins in sewage sludge were first extracted using the processes of ultra-sonication and acid precipitation, and then the humic acid was recovered via membrane filtration. The extraction efficiency was 125.9 mg humic acid?g VSS volatile suspended solids (VSS) and 123.9 mg proteins?g VSS at the optimal ultrasonic density of 1.5 W?mL . FT-IR spectrum results indicated that the recovered proteins and humic acid showed similar chemical characteristic to the natural proteins and humic acid. The acidic solution (pH 2) could be recycled and used more than 10 times during the co-recovery processes. In addition, the dewatered sludge could be easily biodegraded when the humic acid and proteins are extracted, which was essential for further utilization. These findings are of great significance for recovering valuable nutrient from sewage sludge.

Effect of inhibiting tyrosine kinase Src expression on protein phosphatase 2A and tau phosphorylation

摘要: The aim of this study is to investigate the effect of tyrosine kinase Src on Tyrosine 307(Y307) phosphorylation, protein phosphatase 2A (PP2A) activity, and on tau phosphorylation. Specific Src siRNA was transfected into cultured mouse neuroblastoma N2a cells to inhibit the expression of Src protein, and the phosphorylation levels of PP2A Y307 and tau at different sites, as well as PP2A activity were detected at different time points after siRNA transfection. Twelve hours after siRNA transfection, the protein level of Src was dramatically decreased, with decreased PP2A Y307 phosphorylation. However, the total PP2A protein level was also decreased, together with a decreased PP2A activity. Tau was hyperphosphorylated at the Ser198/199/202 sites. Multiple factors may be involved in the cellular regulation of PP2A activity. Inhibiting Src expression could induce inactivation of PP2A and tau hyperphosphorylation.

Encapsulation of polyethylene glycol in cellulose-based porous capsules for latent heat storage and light-to-thermal

摘要: Phase change materials are potential candidates for the application of latent heat storage. Herein, we fabricated porous capsules as shape-stable materials from cellulose-based polyelectrolyte complex, which were first prepared using cellulose 6-(N-pyridinium)hexanoyl ester as the cationic polyelectrolyte and carboxymethyl cellulose as the anionic polyelectrolyte to encapsulate polyethylene glycol by the vacuum impregnation method. Furthermore, the multi-walled carbon nanotube or graphene oxide, which were separately composited into the polyelectrolytes complex capsules to enhance thermal conductivity and light-to-thermal conversion efficiency. These capsules owned a typical core–shell structure, with an extremely high polyethylene glycol loading up to 34.33 g∙g‒1. After loading of polyethylene glycol, the resulted cellulose-based composite phase change materials exhibited high thermal energy storage ability with the latent heat up to 142.2 J∙g‒1, which was 98.5% of pure polyethylene glycol. Further results showed that the composite phase change materials demonstrated good form-stable property and thermal stability. Moreover, studies involving light-to-thermal conversion determined that composite phase change materials exhibited outstanding light-to-thermal conversion performance. Considering their exceptional comprehensive features, innovative composite phase change materials generated from cellulose presented a highly interesting choice for thermal management and renewable thermal energy storage.

Annexin A2-S100A10 heterotetramer is upregulated by PML/RARα fusion protein and promotes plasminogen-dependent

Aberrant expression of annexin A2-S100A10 heterotetramer (AIIt) associated with PML/RARα fusion protein causes lethal hyperfibrinolysis in acute promyelocytic leukemia (APL), but the mechanism is unclear. To facilitate the investigation of regulatory association between ANXA2 and promyelocytic leukemia/retinoic acid receptor a (PML/RARα) fusion protein, this work was performed to determine the transcription start site of ANXA2 promoter with rapid amplification of 5′-cDNA ends analysis. Zinc-induced U937/PR9 cells expressed PML/RARα fusion protein, and resultant increases in ANXA2 transcripts and translational expressions of both ANXA2 and S100A10, while S100A10 transcripts remained constitutive. The transactivation of ANXA2 promoter by PML/RARα fusion protein was 3.29±0.13 fold higher than that by control pSG5 vector or wild-type RARα. The overexpression of ANXA2 in U937 transfected with full-length ANXA2 cDNA was associated with increased S100A10 subunit, although S100A10 transcripts remained constitutive. The tPA-dependent initial rate of plasmin generation (IRPG) in zinc-treated U937/PR9 increased by 2.13-fold, and cell invasiveness increased by 27.6%. Antibodies against ANXA2, S100A10, or combination of both all remarkably inhibited the IRPG and invasiveness in U937/PR9 and NB4. Treatment of zinc-induced U937/PR9 or circulating APL blasts with all-trans retinoic acid (ATRA) significantly reduced cell surface ANXA2 and S100A10 and associated reductions in IRPG and invasiveness. Thus, PML/RARα fusion protein transactivated the ANXA2 promoter to upregulate ANXA2 and accumulate S100A10. Increased AIIt promoted IRPG and invasiveness, both of which were partly abolished by antibodies against ANXA2 and S100A10 or by ATRA.

Transmembrane transport of polycyclic aromatic hydrocarbons by bacteria and functional regulation of membrane

摘要: Explaintheadsorption, uptake and transmembrane transport of PAHs by bacteria. Analyze functional regulation of membrane proteins inthe transmembrane transport. Proteomics technology such as iTRAQ labeling was used to access expressed proteins. Single cell analysis technology wereused to study the morphological structure. In recent years, increasing research has been conducted on transmembrane transport processes and the mechanisms behind the microbial breakdown of polycyclic aromatic hydrocarbons (PAHs), including the role of membrane proteins in transmembrane transport and the mode of transmission. This article explains the adsorption, uptake and transmembrane transport of PAHs by bacteria, the regulation of membrane protein function during the transmembrane transport. There are three different regulation mechanisms for uptake, depending on the state and size of the oil droplets relative to the size of the microbial cells, which are (i) direct adhesion, (ii) emulsification and pseudosolubilization, and (iii) interfacial uptake. Furthermore, two main transmembrane transport modes are introduced, which are (i) active transport and (ii) passive uptake and active efflux mechanism. Meanwhile, introduce the proteomics and single cell analysis technology used to address these areas of research, such as Isobaric tags for relative and absolute quantitation (iTRAQ) technology and Nano Secondary ion mass spectrometry (Nano-SIMS). Additionally, analyze the changes in morphology and structure and the characteristics of microbial cell membranes in the process of transmembrane transport. Finally, recognize the microscopic mechanism of PAHs biodegradation in terms of cell and membrane proteins are of great theoretical and practical significance for understanding the factors that influence the efficient degradation of PAHs contaminants in soil and for remediating the PAHs contamination in this area with biotechnology.

Beyond bag of latent topics: spatial pyramid matching for scene category recognition

摘要: We propose a heterogeneous, mid-level feature based method for recognizing natural scene categories. The proposed feature introduces spatial information among the latent topics by means of spatial pyramid, while the latent topics are obtained by using probabilistic latent semantic analysis (pLSA) based on the bag-of-words representation. The proposed feature always performs better than standard pLSA because the performance of pLSA is adversely affected in many cases due to the loss of spatial information. By combining various interest point detectors and local region descriptors used in the bag-of-words model, the proposed feature can make further improvement for diverse scene category recognition tasks. We also propose a two-stage framework for multi-class classification. In the first stage, for each of possible detector/descriptor pairs, adaptive boosting classifiers are employed to select the most discriminative topics and further compute posterior probabilities of an unknown image from those selected topics. The second stage uses the prod-max rule to combine information coming from multiple sources and assigns the unknown image to the scene category with the highest ‘final’ posterior probability. Experimental results on three benchmark scene datasets show that the proposed method exceeds most state-of-the-art methods.

Diagnostic criteria of latent autoimmune diabetes in adults (LADA): a review and reflection

摘要: Diabetes has become a major public health problem in China nowadays. There are almost 97 million diabetic patients nationwide. Latent autoimmune diabetes in adults (LADA) is a subtype of autoimmune diabetes. Although it has been reported for about 20 years, the diagnostic criteria of this disease remain controversial. The discussion mainly focused on serum autoantibodies, period of insulin need and age of diagnosis. Besides, β cell function, metabolic parameters, genetic factors and cell immunity may also contribute to the formulation of the criteria. Here, we aim to review and discuss the diagnostic criteria of latent autoimmune diabetes in adults.

在疾病干预的众多膜蛋白靶标中,G蛋白偶联受体(GPCR)作为人体内最大的膜受体蛋白家族,成为很多药物的重要靶点,其次是离子通道、转运蛋白和激酶等。膜蛋白在细胞信号转导和运输中发挥了关键作用,当前药物研发面临的挑战在于进一步发掘此类膜蛋白的潜在靶点的干预价值,开发治疗性抗体药物。鉴于特异性抗体能够识别膜蛋白的灵敏特性,以及随着基因工程技术的进步,对已有抗体进行加工改造可获得适应多个靶点蛋白的特异性抗体。然而,成功分离特异靶向膜蛋白抗体取决于一系列因素。我们更易研制和识别结构简单且具有长片段胞外区的抗体分子,但对于高难度的靶点蛋白,如GPCR和其他复杂膜蛋白往往难以得到具有活性的候选抗体。目前若要开发针对复杂膜蛋白(如GPCR、离子通道、转运蛋白和激酶)的抗体药物,必须从抗原靶点设计、抗体筛选策略、先导抗体优化及药物研发模式方面进行考虑。深入研究靶标膜蛋白的结构有助于推进治疗性抗体药物的开发进程。本文概述了抗体靶向复杂膜蛋白的优势和挑战,以及膜蛋白抗原制备和抗体研发策略的最新进展。
